Structure, Function and Selective Inhibition of ADP-ribosyltransferases
Our main focus is on the development of selective inhibitors of enzymes of the human poly(ADP-ribose) polymerase family, recently renamed Diphtheria toxin-like ADP-ribosyltransferases. These enzymes regulate various signalling events through covalent protein modification – ADP-ribosylation. We develop assays suitable for high-throughput screening of inhibitors and use structural information for more rational design. Protein X-ray crystallography is our main tool used to rationalise structure-activity relationships of compounds and to gain understanding of the structure and function of these multidomain proteins.
